The Drosophila gene minibrain codes for a protein kinase. Mutants with a reduced expression of minibrain are characterised by a strongly reduced brain size and learning and memory deficits. Dyrk1A is the mammalian homologue of minibrain; the encoded protein is the founding member of the family of Dyrk protein kinases. The human DYRK1A gene is implicated in the emergence of the cognitive deficits in Down syndrome, due to its location on chromosome 21 in the so-called Down syndrome critical region (DSCR), its overexpression in Down syndrome individuals and also because Dyrk1A-overexpressing mice show learning and memory deficits. Taken together, these data point to a gene dosage dependent function of Dyrk1A/minibrain in processes that are associated with cognitive functions such as learning and memory. The aim of this study was to elucidate the function of Dyrk1A in the adult mammalian brain. Three aproaches were chosen: First, an analysis of the expression of Dyrk1A and of the intracellular localisation of the respective protein in the adult mouse brain was performed. Dyrk1A is expressed in several brain areas, e.g. in the neocortex, in the hippocampal formation and in the cerebellum. In neurons, Dyrk1A is located either mostly nuclear, or predominantly or even exclusively in the cytoplasm, depending on the region and on the type of neuron. Second, to characterise the gene dosis dependent function of Dyrk1A, it was intended to generate transgenic mice that express a third copy of Dyrk1A, as it is the case in human trisomy 21 (Down syndrome). These transgenic mice should express the third copy of Dyrk1A in a region- and time-specific manner, e.g. in principal neurons in the adult forebrain. This should avoid developmental defects and disturbances of, e.g., the motor system and should allow for the specific analysis of the role of Dyrk1A in learning and memory and in brain regions involved in cognitive processes. A BAC (bacterial artificial chromosome) clone containing the whole Dyrk1A locus was isolated. This BAC was modified by homologous recombination in E. coli (GET recombination) to render it suitable for the generation of transgenic mice. Furthermore, a recombination cassette for a second modification step was constructed to allow for the region- and time-specific expression of the Dyrk1A gene on the BAC. Third, a yeast two-hybrid screen to identify proteins interacting with Dyrk1A was performed. Two of six clones obtained were shown to code for proteins interacting specifically with Dyrk1A: Sept4 (Pnutl2/H5/CDCrel-2), a GTPase of the Septin family, and Arip4, a steroid hormone receptor cofactor with ATP-dependent chromatin-remodelling activity. These interactions were confirmed in mammalian cells by coimmunoprecipitation. By in situ-hybridisation, the coexpression of Dyrk1A with the genes of both interactors was shown in various brain regions. In addition, Dyrk1A and Arip4 were shown to be colocalised in a speckle-like nuclear subcompartment in primary rat hippocampal neurons and in mammalian cell lines. These results point to an interaction of Dyrk1A with Arip4 also in vivo and implicate Dyrk1A in steroid hormone-mediated signalling. Furthermore, an interaction between Dyrk1A and Sept5/CDCrel-1, a close relative of Sept4, was shown. Sept5 is thought to play an inhibitory role in the fusion of synaptic vesicles with the presynaptic membrane and transmitter release. The results of this study contribute to the understanding of the role of Dyrk1A in physiological and pathophysiological contexts. Eventually, this should help (i) to elucidate the mechanisms that lead to mental retardation in Down syndrome and (ii) to develop tools to interfere with the function of Dyrk1A and to establish novel strategies for a therapy to alleviate mental retardation in Down syndrome.

The simple metazoan Hydra does posses two genes for caspases: Caspase 3A and caspase 3B. We generated antibodies against these caspases. By immunofluorescence staining and cell fractionation we demonstrated that both caspases are localized in mitochondria in vivo.Caspase 3A is a constitutively active enzyme independent of PCD. After induction of apoptosis caspase 3B translocates from the mitochondria to the cytoplasm and is activated. Activity of 3B is DEVD-specific.

N-Acylhomoserinlactones (AHL) are signalling molecules in gram-negative bacteria, which regulate, in a cell density dependent way, important interactive functions. This phenomenon is known as quorum-sensing. This work characterised the microbial ecology of the autoinducer (AHL) producing bacteria Serratia liquefaciens MG1 and Pseudomonas putida IsoF in the rhizosphere of tomato plants. Gfp- and rfp-tagged strains of the AHL producing wiltypes S. liquefaciens MG1 and P. putida IsoF were compared with its AHL-negative mutants, which were unable to produce AHL. Two kinds of plants cultivating systems were used: a defined axenic system and a complex soil system. The characterisation of the root colonisation behaviour was performed using confocal laserscanning microscopy (CLSM) and cell counting of bacteria. Fluorescence in situ hybridisation (FISH) and terminal restrictions fragment length polymorphism (t-RFLP)-techniques were used to examinate shifts of the bacterial population in the rhizosphere on tomato plants. The effective in situ production and spreading of AHL on tomato roots was demonstrated with P. putida IsoF using an AHL-sensor strain P. putida F117 pKR-C12. AHL was produced in effective concentrations in the rhizosphere of tomato plants and influenced the bacterial rhizosphere population. However, the AHL-production had no influence on the colonization behaviour of the AHL-producing strains S. liquefaciens MG1 and P. putida IsoF.

The endothelium is among the largest organs in the body. Stimuli originating from the blood or from neighbouring cells, like inflammatory cytokines (IC), lead to structural and functional alterations of vascular endothelial cells (EC). These alterations are often referred to as “EC activation”. Activated EC play a key role in different physiological processes like during immune response, in menstruation and in pathological processes like inflammation, allergy, viral infections, atherosclerosis and tumour angiogenesis. The human guanylate binding protein-1 (GBP-1) is a protein of the family of large GTPases. GBP-1 is characterized by a high turnover GTPase activity. Previous work showed that GBP-1 mRNA expression is induced by IC in EC and that GBP-1 is the specific mediator of the anti-proliferative effect of IC on EC in vitro. The main goals of this work were first, to investigate whether GBP-1 may be a molecular marker of IC-activated EC at the protein level in vitro. Second, to investigate GBP-1 expression in human healthy and/or disease tissues and to determine whether GBP-1 may be a molecular marker of IC-activated EC in vivo. To this goal mono- and poly-clonal antibodies against GBP-1 were generated. In vitro studies showed that GBP-1 expression in EC is induced by IFN-, IFN-, IL-1, IL-1 or TNF- but not by other cytokines, chemokines or growth factors. Moreover, simultaneous addition of bFGF and VEGF and IC reduced the IC-induced GBP-1 expression. This indicated that GBP-1 characterizes cells that are preferentially exposed to IC. In vivo studies using immunohistochemistry and immunofluorescence showed that GBP-1 expression is highly associated with vascular EC in a broad range of human tissues. This was confirmed by the simultaneous detection of GBP-1 and the EC-associated marker CD31. Notably, GBP-1 expression was undetectable in healthy skin. In contrast, GBP-1 was highly expressed in vessels of skin diseases with a high inflammatory component including psoriasis, adverse drug reactions and Kaposi’s sarcoma. This indicated that GBP-1 characterizes IC-activated EC in vivo. Further immunohistochemical studies on Kaposi’s sarcoma demonstrated that GBP-1 expression and EC cell proliferation are inversely related. This indicated that GBP-1 may also mediate the anti-proliferative effect of IC on EC in vivo. Finally, GBP-1 was found to be secreted by EC stimulated with IFN- and IFN- in vitro. This finding was confirmed by immunoprecipitation of GBP-1 from cell culture supernatants and by a novel ELISA developed for the detection of GBP-1 in solution. Further characterization of the mechanism of secretion demonstrated that GBP-1 release is due to an 3 Summary energy-dependent mechanism and is not due to cell death. Most importantly, circulating GBP-1 could be detected in increased concentrations in the blood of patients that were subjected to IFN–-therapy or in patients with inflammatory diseases. These findings indicated that GBP-1 is a novel marker of inflammatory vessel activation. Specifically, the serological detection of GBP-1 may open new perspectives for the early detection of inflammatory activation of EC in patients with inflammatory diseases.

MyoD is one of the MRFs (Muscle Regulatory Factors) and it functions to determine the muscle cell fate. The mechanism by which MyoD regulates the muscle development program is very well understood. However, the transcriptional regulation of the MyoD gene itself has not studied in Xenopus. In this thesis, I have analyzed the transcriptional regulatory mechanism of the MyoD gene in Xenopus by different approaches, which include transgenic reporter analysis of its cis-regulatory elements of MyoD transcription and gain-of-function and loss-of-function tests of several potential regulaters. Here I showed that the expression of the XmyoD gene is controlled by a combination of induction, repression and maintenance. One activating motif, one repressing motif and one maintenance motif were found by transgenic reporter analysis. XSRF (Xenopus serum response factor) binds with the maintenance enhancer to maintain the expression of XmyoD gene. A repetitive DNA sequence was discovered in the -2.8/-2.0kb region in the XmyoD genomic sequence. The repetitive DNA sequence in the XmyoD gene locus may produce sense and anti-sense transcripts. In addition, several potential regulators have been analyzed. It has been shown that XSEB-4, a direct target of XmyoD protein, is able to induce the expression of XmyoD gene. Xenopus Ying Yang 1 (XYY1) can repress the expression of the XmyoD gene. Lef-1 is necessary and sufficient for the expression of XmyoD gene. This provides strong evidence that Lef-1 is the transcription factor of the zygotic Wnt signaling pathway that activates the expression of the XmyoD gene.

Uncovering the motifs of a higher order nuclear architecture and its implications on nuclear function has raised increasing interest in the past decade. The nucleus of higher eukaryotes is considered to display a highly dynamic interaction of DNA and protein factors. There is an emerging view that there are hierarchical levels of gene regulation, reaching from epigenetic modifications at the DNA- and histone level to a higher order functional nuclear topology, in the context of which gene-activating and -repressing processes influence the gene expression profile of an individual cell beyond the sequence information of the DNA. The present work focuses on the analysis of the dynamic aspects of higher order nuclear architecture in living cells. As a prerequisite, an in vivo replication labeling strategy was developed, that enabled the simultaneous visualization of early and mid-to-late replicating chromatin as well as single chromosome territories on the basis of a labeling/segregation approach. The presented scratch replication labeling protocol combines a high labeling efficiency with reduced “damaging” effects and can be successfully applied to a number of adherently growing cell lines, including primary human fibroblasts. In addition, a live cell observation system was developed that facilitates time-lapse confocal (4D) microscopy over elongated time periods which made it possible to follow a complete cell cycle or more. To address possible long-range movements of chromosome territories (CTs) during an entire interphase, fluorescence labeling of a small number of CTs was performed in living HeLa cells stably expressing histone H2B-GFP. This was achieved by in vivo scratch replication labeling with fluorescent nucleotides. Labeled cells were cultivated for several cell cycles until labeled chromatids had segregated. Such cells were followed by time-lapse confocal microscopy over time-scales of up to 20 hours covering major parts or the complete cell cycle. Positional changes of the intensity gravity centers of labeled CTs in the order of several µm were observed in early G1, thereafter, the positions remained within a range of ~1 µm till the end of G2. In conclusion, CT arrangements were highly constrained from mid G1 to late G2 / early prophase, whereas major changes of CT neighborhoods occurred from one cell cycle to the next. More extended movements observed in early G1 might play a role when CTs “home in” to establish a non-random radial CT arrangement. To analyze possible changes of chromosome arrangements from one cell cycle to the next, nuclei were photobleached in G2 maintaining a contiguous zone of unbleached chromatin at one nuclear pole. This zone was stably preserved until the onset of prophase whereas unbleached chromosome segments were often observed to become located at distant sites in the metaphase plates. Accordingly, chromatin patterns observed in daughter nuclei differed significantly from the mother cell nucleus, indicating that CT neighborhoods were not preserved during mitosis. The variability of CT neighborhoods during clonal growth was further confirmed by 3D-FISH experiments. A series of experiments of a more preliminary character looked at the influence of the nuclear lamina in constraining and determining a higher order nuclear architecture by selectively interacting with mid-to-late replicating chromatin. Simultaneous immunodetection of lamin B on two-color replication labeled neuroblastoma cell nuclei revealed specific attachment of the mid-to-late replicating chromatin compartment not only along the periphery but also inside the nucleus along invaginations of the lamina. 4D-live cell observation of lamin C-GFP expressing CHO cells with mid-to-late replicating chromatin labeled simultaneously revealed concomitant movements of replication foci attached to lamin invaginations. Moreover, a functional essay was employed which uses injection of a dominant negative lamin A mutant protein (ΔNLA) to cause a reversible disruption of the nuclear lamina. Initial results point to concomitant distortion of the mid-to-late replication pattern and a preferential attachment of the respective chromatin sites to partially disrupted lamin B (as compared to lamin A and nuclear pore complex). Finally, a model is presented on the chromosome positioning in mammalian nuclei depending on cell cycle and nuclear shape.

During my PhD thesis I was working on the identification of novel apoptosis-inducing genes by using a novel genetic expression screen (Grimm and Leder, 1997). One of the identified genes turned out to be a evolutionary conserved cDNA that codes for a novel BH3-only protein of 219 amino acid residues which was named Spike, for Small protein with inherent killing effect. Spike was then in the course of my PhD thesis extensively characterised, molecularly and biochemically. In summary, upon overexpression in mammalian cells Spike efficiently leads to all features of apoptosis, such as phenotypic alterations, cytochrome c release, caspase activation and DNA degradation. It was shown that Spike localised to the endoplasmic reticulum, where it interacts with a recently identified apoptosis regulating protein complex, consisting of Bap31, Bcl-2, Bcl-XL and an ER-specific isoform of caspase-8: pro-caspase 8L (Breckenridge et al. 2002). Although no direct interaction with anti-apoptotic members of the Bcl2-family could be observed, the importance of the BH3-like sequence for the apoptosis-inducing activity of Spike was demonstrated by using point mutations of conserved amino acid residues of this motif (Mund et al. 2003). Instead of directly interacting with anti-apoptotic members of the Bcl-2 family Spike is able to interfere with the complex formation between Bap31 and Bcl-XL. Based on these data I proposed a model according to which the complex on Bap31 is controlled by Bcl-2/Bcl-XL as long as they are associated. Displacement of Bcl-2/Bcl-XL from Bap31 by Spike leads to the formation of a pro-apoptotic complex. In addition, Spike appears to be implicated in the cell death signal of the Fas receptor. I observed that a dominant-negative version of Spike and a highly effective anti-sense oligonucleotide significantly reduced Fas-mediated DNA fragmentation whereas no reduction was detected in TNFa-induced cell death (Mund et al. 2003).
