Percutaneous coronary intervention (PCI) is the most common treatment for coronary artery disease (CAD). The first form of PCI introduced was balloon angioplasty. After that, the advent of coronary stents (tubular wire mesh for intravascular mechanical support) led to a new era in interventional cardiology. Through the implantation of bare metal stents (BMS), all three limitations of balloon angioplasty – coronary artery dissection, elastic recoil and negative remodelling – are prevented. Unfortunately, bare metal stents have their own drawbacks: most significantly, in-stent stenosis as a result of deep focal vascular injury caused by stent struts, followed by excessive tissue proliferation. This drawback has since been addressed with the introduction of drug eluting stents (DES). Both BMS and DES are permanent stents. As foreign bodies implanted into coronary vessels, they cause the following adverse effects: hypersensitivity reaction, chronic inflammation, elimination of vasomotion, and stent thrombosis. Biodegradable stents have been set forth as a candidate to overcome the drawbacks of permanent stents through providing temporary mechanical stability for a vulnerable lesion before complete degradation without long-term impairment of vessel function. The two main materials used in biodegradable stents are poly-L-lactide and metal (or AMS, short for absorbable metal stents). One type of AMS is magnesium alloy. Before AMS become a standard in the treatment of CAD, more research is required to better understand their degradation kinetics and mechanical stability. In order to complete these studies, however, the stents must provide adequate opacity for visualization – wherein lies the challenge. It is not possible to visualize magnesium stents with coronary angiography. Newer imaging modalities such as optical coherence tomography (OCT) and intravascular ultrasound (IVUS) have been proposed as techniques to visualize these stents and their biodegradation. The aim of this study was twofold: first, to study the available in-vivo visualization techniques (OCT and IVUS) in order to identify their strengths and weaknesses in assessing the biodegradation process of AMS through comparison with histology; and secondly, to identify a new histological technique for studying the distribution of magnesium and its degradation products into surrounding tissue upon biodegradation. Four Gottingen mini pigs were implanted with AMS and BMS, and assessed with IVUS and OCT under fluoroscopic guidance at the time of implantation and prior to explantation (4 weeks later). Upon completion of the in-vivo studies, the hearts of the study objects were harvested for histological processing. Results showed that both IVUS and OCT are effective visualization techniques in studying the biodegradation process of AMS. IVUS is superior to OCT in capturing vessel morphometry thanks to its ease of use and consistently high image quality, thus enabling us to study vessel dimensions during biodegradation. OCT, however, is a better technique for detailed vessel assessment thanks to its higher resolution, and helps us to detect qualitative changes during biodegradation. The two methods correlate moderately during the morphometric analysis. The histological studies on the other hand showed a poorer correlation with the in-vivo techniques. This was likely due to strut integrity compromised during cryosectioning and subsequently washed away after staining, as in an adjunctive study we were able to show that staining did not affect morphometry. The measurements were consistently largest with IVUS, and the smallest with histology (IVUS > OCT > histology). Though IVUS and OCT together offer a gross understanding of AMS biodegradation, in order to complete this view at a cellular level one must employ a third technique. The technique investigated in this study was titan yellow staining, which proved to be a feasible method for capturing the biodegradation process and the distribution of magnesium within the vessel wall. This study showed for the first time that biodegradable magnesium stents and their degradation products can be visualized effectively ex-vivo, and that analysis of these images will allow us to better understand the changes due to degradation during in-vivo visualization. Its simplicity and speed make titan yellow preferred to the in-vitro corrosion techniques previously employed. The procedure, however, must be further modified in order to improve its effectiveness over a longer period of time. Once this is achieved, it will then be possible to measure the density of degradation products away from struts over time, so as to more thoroughly understand the degradation kinetics.

Lung cancer is the leading cause of cancer deaths worldwide. Despite advances and progresses in surgery, chemotherapy, and radiotherapy over the last decades, the death rate from lung cancer has remained largely unchanged, which is mainly due to metastatic disease and multi drug resistance. Because of the overall poor prognosis, new treatment strategies for lung cancer patients are urgently needed. The aim of this study was to investigate the interactions between pemetrexed and vinorelbine for human adenocarcinoma via various chemotherapy schedules. Vinorelbine and pemetrexed caused a strong dose-dependent cytotoxic effect in both HCC and cisplatin resistant HCC (HCC-res) cells. The IC50 values of vinorelbine against HCC and HCC-res cells were 10.34±1.12 nM and 9.98±2.12 nM, respectively. The IC50 values of Pemetrexed against these cells were 110.77±17.28 nM and 118.89±18.77 nM respectively. The application of different therapy schedules induced a significant time dependent cell growth inhibition on HCC naïve and cisplatin resistant cells. The therapy scheme of cisplatinpemetrexedvinorelbine showed the strongest inhibitory effect on both HCC and HCC-res cells. The application of different therapy schedules on HCC and HCC-res cells increased the percentage of cells undergoing apoptosis, except the application of vinorelbine alone. In both HCC and HCC-res cells, cisplatinpemetrexedvinorelbine was found the most effective to induce apoptosis. The application of different therapy schedules on HCC and HCC-res cells increased cytoplasma calcium concentration. Only the application of vinorelbine alone failed to increase calcium concentration in HCC cells. The most elevated calcium concentration was found in the cells treated with cisplatinpemetrexedvinorelbine in both HCC and HCC-res cells As a conclusion, the sequential application of cisplatin, vinorelbine and pemetrexed has a synergistic effect in cell growth inhibition, apoptosis induction, and calcium concentration elevation in HCC and HCC-res cells. The calcium overload could lead to apoptosis, which was related to the cell growth inhibitory effect of chemotherapeutics in lung cancer cells. It might cast a light to develop chemotherapy schedules for patients, and to overcome cisplatin resistance in lung cancer.

Regulatory T-cells (Treg) are important regulators of immune responses. In AML patients before/after immunotherapy (stem-cell-transplantation (SZT) or donor-lymphocyte-infusion (DLI)) their suppressive role can contribute to suppress severe graft-versus-host-reactions (GVHD), but also to impair antileukemic reactions. Aim: Since leukemia-derived dendritic-cells (DCleu) are known to improve antileukemic functionality of T-cells we evaluated the composition and development of distinct Treg-subtypes in AML-pts (n=12) compared to healthy probands (n=5) under unstimulated conditions and during stimulation with DCleu-containing DC (‘DC’) or blast-containing mononuclear cells (‘MNC’) in 0-7 days mixed lymphocyte cultures (MLC) by flowcytometry. T-cell-subgroups in AML pts were correlated with antileukemic functionality before and after ‘DC’ or ’MNC’ stimulation by functional fluorolysis-assays. Results: 1. AML-pts’ T-cells presented with significantly higher frequencies of Treg-subgroups in unstimulated T-cells compared to healthy probands. 2. After 7 days of ‘DC’ or ‘MNC’-stimulation all Treg-subtypes generally increased; significantly higher frequencies of Treg-subtypes were still found in AML-pts. 3. Antileukemic cytotoxicity was achieved in 36% of T-cells after ‘MNC’- compared to 64% after ‘DC’-stimulation. Antileukemic activity after ‘DC’ but not after ‘MNC’-stimulation correlated with significantly lower frequencies of Treg-subtypes (CD8+ Treg/Teff/em reg). Furthermore, cut-off-values for Treg subpopulations could be defined allowing a prediction of antileukemic response. Conclusion: We demonstrate a crucial role of special Treg-subtypes in the mediation of antileukemic functionality. High CD8+Treg, Teff/em reg and CD39+T-Zellen clearly correlated with reduced antileukemic activity of T-cells. ‘DC’ stimulation of T-cells contributes to overcome impaired antileukemic T-cell-reactivity. Refined analyses in the context of clinical responses to immunotherapy and GVHD reactions are required.

Circadian clocks are endogenous cellular mechanisms that control daily rhythms of physiology and behaviour. The adjustment of the circadian clock to the 24 h period of a day is commonly accomplished by several environmental cues, e.g. temperature, light and nutrition. For one light input pathway the mechanism that synchronises or entrains Neurospora’s clock is supposed to be known. Nevertheless, there are plenty more environmental cues that have an obvious impact on the circadian clock, e.g. temperature. The environmental cue “temperature” was underrepresented in studies about Neurospora’s circadian clock, while several clock studies focused on stationary conditions rather than changing ones. As a result, the functionality and adaptation of circadian clocks were underestimated, and thus new clock components could be overlooked due to screening on constant darkness. It was therefore important to develop a novel strategy in screening mutants that challenged the circadian clock of Neurospora crassa entirely on temperature alternations. A temperature cycle of low amplitude (22° C cold and 27° C warm) and of short period (8 h cold and 8 h warm) applied as Zeitgeber stimulus. A mutant library was created with insertional mutagenesis via electroporation in order to transform a BASTA resistance gene into conidial nuclei. As a consequence, a novel method of rescuing the mutations was established, which combined the process of mapping, partial cloning and PCR and was called Size Selected Fragment Plasmid Rescue or SSFPR in short. During the screening of several hundred mutants among the novel protocol, a known clock gene, frequency, was identified and characterised. The identification of several mutants with altered clock phenotypes has on one hand confirmed the general approach of this study and on the other proved that the greater sensitivity for the temperature screen can bee used to detect mutant phenotypes.
