Aim of the present study was, to show relations between various metabolic parameters and the occurrence of inflammatory diseases of the uterus in dairy cows. Additionally, we wanted to find limiting values that show an increased risk of disease if they were exceeded or fallen short of. In eight dairy herds 124 cows and heifers were examined weekly from week 2 before calving to week 5 postpartum. Metabolic parameters in blood as total protein, albumin, total bilirubin, aspartate aminotransferase, γ-glutamyl transferase, glutamate dehydrogenase and β-hydroxy butyric acid were examined once weekly from week 2 before to week 2 after parturition. Parameters in milk such as fat, lactose, fat-/lactose-quotient, protein, fat-/protein-quotient and urea were examined or calculated in week 1 and 2 postpartum in cisternal and residual milk, body condition was determined with body condition scoring on every date of examination. Gynaecologic examinations were conducted in week 2, 4 and 5 postpartum. Furthermore, additional data as milk yield, calving interval, dead births, problems at parturition, as well as the time from parturition to first insemination and calving to conception interval, was documented. For the evaluation of the data, cows were divided into two groups. Cows, that suffered from puerperal metritis, clinical metritis, clinical endometritis or pyometra were assigned to the group “ill”. Cows, that didn’t suffer from any uterine disease on one of the dates of examination, were assigned to the group “healthy”. If there were significant differences in parameters between groups, we formed limiting values for these parameters. The exceeding or deceeding of these limiting values showed a higher risk of disease. Probably the energy deficit, which is reflected in the change of metabolic parameters, is most likely responsible for an impairment of the immune system and thereby for the higher risk of suffering uterine disease.

≥ 7,05 µmol/l (Sensitivität (Sens.) 53%, Spezifität (Spez.) 83%, Odds ratio 5,7), β- HBA an Tag 1 p.p.≥ 0,68 mmol/l (Sens. 53%, Spez. 79%, Odds ratio 4,4), Albumin an Tag 1 p.p.≥ 34,35 g/l (Sens. 50%, Spez. 72% Odds ratio 2,7) sowie an Tag 11 p.p. < 31,85 g/l (Sens. 81%, Spez. 79%, Odds ratio 16,8), AST an Tag 1 p.p. ≥ 80,8 U/l (Sens. 41%, Spez. 86%, Odds ratio 4,3) und an Tag 11 p.p. ≥ 117,8 U/l (Sens. 47%, Spez. 84%, Odds ratio 4,5).

SUMMARY The scope of the present work was to provide an scientific overview of the actually state of canine genetics of hereditary diseases. Causative mutations, inheritance modes, breed disposition and incidence of the single genetic defects in the affected dog populations have been outlined. Additionally a special focus aims on the gathering of actually available genetic tests for respective hereditary diseases within different dog breeds. The present state of science covers 76 genetically characterized hereditary diseases of the dog (status: January, 2009). Their inheritance mode, genetic basis, pathophysiology, breed dispositions and genetic control are shown here in detail. Up to date, 61 DNA tests are available for canine hereditary diseases. A compendium of the actually available genetic tests for the affected breeds with different suppliers is included in the appendix of this work. In the sector of genome analysis and screening tests for dogs, high research activity can be observed. This contributes to the specific suitability of the species dog as an animal model for the investigation of human hereditary diseases. The partial considerable inbreeding of some dog breeds leads to the appearance of numerous monogenetic hereditary diseases which are mostly of recessive nature. In addition, research results of the last few years demonstrate that these diseases are phenotypical or even genotypical homologues to human hereditary diseases. The number of the dog´s identified genetic mutations, which are connected to hereditary diseases, has increased in the past years and will increase continuously in the next years. An application of the enlarged genetic research results is as well of interest to the human medicine, as to the stockbreeding. The application of molecular-genetic screening methods offers dog breeders the possibility to determine the genetic status of their breeding dogs, to select healthy animals and to minimize future hereditary defects. The incidence of hereditary diseases can be reduced using genetic screening tests on the breeding dogs before breeding, introducing DNA of breeding dogs from other populations and avoiding the inbreeding within different pedigrees of dogs. Since a couple of years numerous screening and breeding programmes and the generation of genotype data banks are an obligation within many associations of dog breeders. This might accelerate an offbreeding of hereditary defects. The rising quantity of genetic screening tests and further investigation of undecoded mutations, will allow researches to amplify the understanding in the wide field of the polygene diseases. Relieving the succeeding generations of pedigree dogs from monogene and polygene hereditary sufferings is the future aim.

Introduction of the RCAS retroviral gene transfer technology for functional genomic studies in the chicken The recently sequenced chicken genome now allows the identification of so far unknown genes. Some of them may play a potential role in the regulation of the immune system. In future studies roles of these genes have to be analyzed. At this in vitro studies will be used firstly. But in vivo functions of those genes will have to be described in the end. Therefore techniques for a fast and efficient over expression and knock down of candidate genes have to be established. Goal of this work was to prove the applicability of the RCAS retroviral vector system, which has been developed in the 1980s. This highly efficient system is based on a replication competent Rous Sarkoma-Virus and has already been used in cell and developmental biology studies in the chicken. However studies on its application after hatch haven´t been performed so far, despite its particular relevance for studies on the chicken immune system as it is widely immature at the time of hatch. In the first part of this work, a green fluorescent protein (GFP) expressing vector was constructed. An influence of the chicken line on the susceptibility of chicken embryo fibroblasts was detected in flow cytometric analyses. A possible reason could be the presence of endogenous retroviruses in less susceptible chicken lines leading to receptor interference. In accordance with this observation in in vivo tests retrovirus free chickens showed more homogenous plasma titers of RCAS based protein than a commercial chicken line. As GFP expression was predominantly seen in endothelial cells, the RCAS technology can be considered as particularly eligible for the expression of soluble proteins. Additionally viral protein was demonstrated to be expressed in bursal cells and heart muscle. For further characterization of the RCAS system vectors for the expression of the cytokine chBAFF or a soluble chBAFF receptor construct (huBCMA-Fc) were used. Plasma expression of biologically active cytokine was detectable throughout the entire experiment (two months). RCAS(BP)A-chBAFF-Flag transduced birds showed in comparison to controls significantly increased B cell frequencies in the spleen as well as enhanced plasma antibody titers. On the other hand animals expressing the soluble receptor huBCMA-Fc showed a significant reduction of splenic B cell frequency and an inhibited development of the bursa of fabricius. These findings correspond to the data obtained in studies with recombinant proteins. In the second part of this work the experiences with the RCAS technology were used to evaluate the function of the CD40/CD40L system as a second cytokine system. A soluble chCD40-huFc fusion protein was constructed as a functional inhibitor for CD40/CD40L interaction. In vitro tests showed that chCD40-huFc was able to bind a soluble chCD40L fusion protein. In vivo the RCAS based expression of chCD40-huFc caused a reduction of the blood B cell frequency and the bursa weight. Possibly the functional inhibition of the CD40/CD40L system decreases the number of B cells emigrating from the bursal medulla. Unlike in humans with a defect in the CD40L gene in chickens expressing chCD40-huFc the plasma IgM titer was not increased. However plasma titers of IgA and IgG were decreased, comparably with humans and mice with CD40L defects. The RCAS gene transfer technology was proved in two different model systems as an excellent system for analyses of cytokine functions in the chicken. Furthermore this system could also be used for functional analyses of growth factors and hormones.

This dissertation focuses on epidemics which affected horses of the Bavarian army between 1880 and 1920. It is based on files found in the war archive in Munich and supplemented by contemporaneous professional literature when the documents were incomplete. These files show that contagious pneumonia (synonym: Pleuro-Pneumonia Contagiosa Equorum) proved to be a great problem at the end of the 19th century, and there was no solution initially. The discovery of salvarsan by Paul Ehrlich in 1909 and its use in equine medicine three years later proved to be an effective therapy against this frightening epidemic. Although occasional cases occurred during World War I, the disease was controlled and did not limit the marching ability of the troops. It was a different situation with glanders. Due to strict precautions, horses of the Bavarian army were not affected by this epizootic disease during peacetime. Yet, once the army entered into World War I, the disease flared up and threatened the operation of the troops. Systematic use of the Mallein test and serological blood examination led to quick containment. Even though the disease was not completely eradicated, the operations of the troops were not endangered anymore. The Bavarian army horses were absolutely free of mange before entering World War I. However, the situation changed considerably a few months after the war began. The mange epidemic hit a large number of horses and the known therapies did not cure them. The marching ability of the troops was soon at serious risk. A breakthrough was finally discovered by exposing the horses’ bodies to vaporized sulfuric acid. However, this breakthrough came too late to have deep impact on the course of the war. In the post-war period only glanders and mange were a risk for the horse of the Bavarian army. In Germany, glanders were finally eradicated in 1955.

The goal of this study was to examine the suitability of five disease agents that are essential in swine production, in a health monitoring program. Not only the selected disease agents and the diagnostic methods were examined but also the sample size. Four market trading companies from Lower Saxony and Schleswig-Holstein provided 38 farrowing farms for the study, and sampling was done on a quarterly basis. Fifteen blood samples were taken from sows every time and serologically examined for Salmonella and in one quarter only PCV2 was tested. A total of fifteen blood samples from feeder pigs were serologically examined for Salmonella in the first and second quarter. In the course of the following testings, instead of blood probes 20 faecal samples were pooled to create four samples which were bacteriologically examined. Additionally, 30 blood probes from three growing stages were examined for antibodies against PCV2 using Ingezim PCV IgG/IgM ELISA. Twenty nasal swabs were pooled to create four samples that were tested for the EU and US strains of PRRSV. Furthermore, the four pooled faecal samples were examined for Brachyspira spp. using Multiplex-PCR, and the probes from the first two quarters were also examined for Campylobacter spp.. The farms were assigned into one of four categories, 0 to 3, based on their results from the serological examination for Salmonella and similar to those of the Salmonella VO and QS. Based on the sow results, 10.8% of the farms were in the category 0, 67% in the category 1 and the rest of the farms fell into the categories 2 and 3. Feeder pigs from three farms were placed in the category 1 and the rest were assigned the category 0. In the following bacteriological examination, Salmonella was identified in ten farms. The bacteriological examination method was better in the flatdeck when compared to the serological method. The serological examination of the sows proved to be effective for a health monitoring program. Antibodies for PCV2 were found in the sows in all farms, which confirms its ubiquitous distribution. No seroconversion could be identified in the weaner-to-feeder pigs in 10 farms. Many farms began piglet vaccination during the project. It was noted that the vaccination influenced the results of the Ingezim ELISA in these farms and that an examination is not necessary. This test can be well implemented in a monitoring program in farms that do not vaccinate. However, this test also identifies maternal antibodies and this needs to be taken into consideration when interpreting results. B. hyodysenteriae could not be identified in any farms. Other Brachyspira species that occur in swine could not be identified with the exception of B. innocens that was found in two farms. PCR appears to be a suitable diagnostic tool for a health monitoring program. The examination for Campylobacter spp. showed a wider distribution of C. coli, and 89.4% of the farms tested positive. C. jejuni could only be found as a coinfection with C. coli in one farm. In four farms, no Campylobacter sp. could be identified. In this case, PCR is also suitable for examining faecal samples. The outcome of the PRRSV examination showed highly differing results in every test period. 78.9 % of the farms altered their status during the project. This infers that the sample size was inadequate in order to determine the actual status of the farm. Moreover, a significant correlation between vaccine strain and the PCR results was observed. The PCR only differentiated between both genotypes but not between field and vaccine strains. However, a differentiation is advisable for a health monitoring program. Alternatively, the examination for PRRSV-free nucleus farms could be restricted and simply the vaccine status could be documented in other production units.
