The Mediator of transcriptional regulation is the central coactivator that enables a response of RNA polymerase II (Pol II) to activators and repressors. Yeast Mediator has a size of more than one MDa and consists of 25 different polypeptides. Biochemical studies defined three Mediator modules in yeast, the head (MED17) the middle (MED9/MED10) and the tail (MED15) modules. During this work, an E.coli coexpression-copurification system was developed, which allowed to study pairwise interactions of Mediator middle module subunits. With the help of this system I reconstituted a complex of two essential and conserved yeast Mediator middle module proteins, the MED7/MED21 heterodimer, and solved its crystal structure. The heterodimer forms an extended structure, which spans one third of the Mediator length, and almost the diameter of Pol II. It shows a four helix bundle and a coiled-coil protrusion connected by a flexible hinge. Multiple conserved patches can be identified on the surface, which allow for assembly of the middle module. A combination of the coexpression-copurification system and assembly of subcomplexes allowed the reconstitution of a five-subunit Mediator middle module subcomplex. The reconstituted subcomplex is able to bind Pol II in vitro. MED6 associates with the middle module and forms a bridge to the head module. The potential flexibility of this bridge and the MED7/MED21 hinge can account for changes in Mediator structure upon its binding to Pol II or to activators.

Flt3 is the most recently discovered member of the platelet-derived growth factor receptor (PDGFR) subfamily of receptor tyrosine kinases (RTKs). It is expressed on early hematopoietic progenitor cells and is involved in their growth. Activating mutations of Flt3, such as internal tandem duplications (ITD) in the juxtamembrane (JM) region of Flt3, are among the most common genetic alterations found in patients with acute myeloid leukemia (AML). For this reason Flt3 is an attractive target for specific tyrosine kinase inhibitors, and a number of such compounds are currently being tested in clinical trials. However, the mechanisms of Flt3-mediated signal transduction are poorly understood. Src family tyrosine kinases (SFKs) are involved in the signaling processes of various RTKs, including those of the PDGFR subfamily . Most SFKs are expressed predominantly or exclusively in hematopoietic cells, and at least Hck and Lyn were found to be activated in AML patients. These observations indicate that SFKs could also be involved in Flt3 signaling. In this work the role of SFKs in signal transduction of the Flt3 receptor was investigated. A panel of Hck and Flt3 mutants was used in biochemical and biological assays to deduce the contribution of kinase activity, SH2 and SH3 binding domains and tyrosine phosphorylation status to Flt3-SFK interaction. The work presented shows that SFKs interfere with the maturation of wild type and Flt3 ITD receptors in human embryonic kidney 293 (HEK- 293) cells in a kinase-dependent manner. The SFK-mediated effect on receptor maturation is reflected by accumulation of the immature, intracellular form of Flt3 at the expense of the mature, plasma membrane-inserted form of the receptor. This effect of SFK kinase activity on receptor maturation is not limited to Flt3, because analogous results were also obtained for the Kit receptor, another member of the PDGFR subfamily. The demonstration of the role of SFKs in regulation of receptor maturation is novel and the exact mechanism underlying this effect requires further investigation. Using the HEK-293 cells it was also shown that Hck is able to phosphorylate Flt3 on tyrosine residues and to associate with autophosphorylated Flt3 in an SH2 domain-dependent manner. Hck-mediated tyrosine phosphorylation occurs in the JM region of Flt3. Tyrosine residues 589 and 591 in the JM region, when phosphorylated, were also identified as the docking sites for Hck. Although Hck phosphorylation and binding sites on Flt3 overlap, Hck-mediated phosphorylation of Flt3 is not sufficient for Flt3-Hck association. In contrast to the situation in HEK-293 cells, Hck neither interferes with Flt3 maturation nor phosphorylates Flt3 on tyrosine residues, and does not detectably associate with the Flt3 receptor in the hematopoietic murine cell line 32D clone 3 (32D cl.3). However, the level of the ectopic expression of Hck in 32D cl.3 cells was lower than in HEK-293 cells. This difference in Hck expression level probably accounts for both the lack of the Hck-mediated phosphorylation of Flt3 and the interference with its maturation in 32D cl.3 cells. The lower Hck expression level, however, does not account for the lack of detectable association of Hck with Flt3 in 32D cl.3 cells. Using biological assays in which activation of Flt3 can partially overcome the requirement for cytokine stimulation in 32D cl.3 survival and proliferation, it was shown that Hck is not involved in Flt3 signal transduction leading to Flt3-mediated cell survival and Flt3 ITD-dependent cell growth. Therefore, biological relevance of SFKs in Flt3 signaling remains unclear. In summary, although the binding and phosphorylation of Flt3 by Hck can be shown, the biological relevance of Hck in Flt3 signaling remains to be formally demonstrated. This research led to the novel finding that, at least when over-expressed or hyperactivated, SFKs interfere with the maturation process of RTKs. Various studies have shown that the ligandindependent activation of RTKs as well as their premature phosphorylation can interfere with maturation of RTKs. This study reveals that, by virtue of their phosphorylating of Flt3, Kit and possibly other RTKs, SFKs can regulate maturation of these RTKs and consequently alter their transport to the plasma membrane. Further studies are required to investigate the biological relevance of this function of SFKs.

This thesis focuses on the experimental and theoretical investigation of small molecules containing phosphorus, nitrogen and sulfur. The first part of this work presents synthesis and characterisation of molecules bearing a P-N-N-group: N,N´,N´-[tris(trimethylsilyl)]-hydrazino-diphenylphosphane, (TMS)2N-(TMS)N-PPh2, and N,N´,N´-[tris(trimethylsilyl)]-hydrazino-phenyl(chloro)phosphane, (TMS)2N-(TMS)N-P(Cl)Ph, were obtained in the reaction of bis-[lithiumtris(trimethylsilyl)hydrazide] with PhnPCl3 - n (n = 1, 2). The structure and bonding of both species are discussed on the basis of experimentally observed (X-ray, Raman, NMR, and MS) and theoretically obtained data (B3LYP/6-31G(d,p), NBO analysis). Oxidation with sulfur and selenium results in the formation of (TMS)2N-(TMS)N-P(S)-Ph2, (TMS)2N-(TMS)N-P(Se)Ph2, (TMS)2N-(TMS)N-P(S)Ph(Cl) and (TMS)2N-(TMS)N-P(Se)Ph(Cl). Moreover, the thermal decomposition of N,N´,N´-[tris(trimethylsilyl)]hydrazine-dichlorophosphane, (TMS)2N-(TMS)N-PCl2 and the reaction with magnesium have been investigated. The formation and molecular structure of the novel MgCl2(THF)2* 2Mg[(TMS)NP(O)2N(TMS)2](THF) salt containing the hitherto unknown (TMS)NP-(O)2N(TMS)22- anion are discussed. DFT calculations (B3LYP/6-311+G(3df,2p)//B3LYP/6-31G(d,p)) are used to evaluate the bonding, ground-state structures, and energy landscape for the different isomers of (TMS)2N-(TMS)N-PCl2: the thermodynamics and kinetics of the successive elimination of chlorotrimethylsilane (TMS-Cl) resulting in the formation of covalent azide analogues such as TMS-PNN or TMS-NNP are discussed. While investigating the reaction of (TMS)2N-(TMS)N-PCl2 with Lewis acides a galliumtrichloride-adduct of 4-bis[trimethylsilylamino]-1,2,4,3,5-triazadiphosphole was discovered. This hitherto unknown triazadiphosphole represents one of the few examples of a five-membered ring-molecule containing only phosphorus and nitrogen. This ring system was unequivocally characterized by X-Ray, Raman, NMR and MS analysis. Furthermore the Staudinger-type reaction of N,N´,N´-[tris(trimethylsilyl)]-hydrazino-diphenylphosphane with SP(N3)3 and and C3N12 in different stoichiometries were investigated. It is possible to substitute only one azide group in SP(N3)3 and up to two azide groups in C3N12 with (TMS)2N-(TMS)N-PPh2, resulting in the expected products. Upon heating of a solution of (TMS)2N-(TMS)N-PPh2 and SP(N3)3 a new eight-membered ring containing phosphorus and nitrogen in alternating order was obtained. The molecule contains a long pnicogene-chain (eleven atoms in total, three phosphorus- and eight nitrogen-atoms). The reactions of (NSCl)3 and NSCl2- with AgX salts (X = CN, SCN, OCN), as well as the reaction of (NSCl)3 with [PPh4]X (X = CN, SCN, OCN) and HgX (X = CN, SCN) have been investigated in a combined experimental and theoretical study. Additionally, the reaction of NSCl2- salts with hydride-transferring-agents like NaBH4 or NaBH3CN was studied. The thermodynamics as well as structure and bonding of the formation of NSX, NSX2- and NSXY- (X, Y = Cl, H, CN, SCN, OCN) have been studied. Unfortunately the preparation of these species did not suceed. The substitution of sulfur by selenium with SeO2, as reported by Rawson, was investigated on the salts of NSCl2- and S2N3+. These reactions resulted polymeric products and selenium; the formation of a nitrogen-selenium-species could not be confirmed.

The Mediator of transcriptional regulation is the central coactivator that enables a response of RNA polymerase II to activators and repressors. It is conserved from yeast to human and consists of 25 subunits in yeast that are organized in four modules called head, middle, tail, and CDK8/Cyclin C module. Despite its central role in transcription the functional mechanism remains enigmatic. To overcome the lack of detailed structural data on the Mediator a recombinant expression system was established that allows large-scale purifications of Mediator head module subcomplexes. It has been shown that via limited proteolysis assays and multicistronic expression the problems of insolubility and low expression rates of Mediator subunits can be overcome, paving the way for structural studies on subcomplexes of the Mediator head module. First data indicated that a reconstitution of the complete head module is within close reach. Large-scale copurification data led to a detailed interaction map of subunits and subcomplexes from within the head module and towards the middle module. The second part of this work describes the structure solution of a subunit in the CDK8/Cyclin C module – Cyclin C. Cyclin C binds the cyclin-dependent kinases CDK8 and CDK3, which regulate mRNA transcription and the cell cycle, respectively. The crystal structure of Cyclin C reveals two canonical five-helix repeats and a specific N-terminal helix. In contrast to other cyclins, the N-terminal helix is short, mobile, and in an exposed position that allows for interactions with proteins other than the CDKs. A model of the CDK8/Cyclin C pair reveals two regions in the interface with apparently distinct roles. A conserved region explains promiscuous binding of cyclin C to CDK8 and CDK3, and a non-conserved region may be responsible for discrimination of CDK8 against other CDKs involved in transcription. A conserved and Cyclin C-specific surface groove may recruit substrates near the CDK8 active site. Activation of CDKs generally involves phosphorylation of a loop at a threonine residue. In CDK8, this loop is longer and the threonine is absent suggesting an alternative mechanism of activation is discussed based on a CDK8-Cyclin C model.

Dense collagen implants were developed which can be easily manufactured by extrusion at room temperature without the need of organic solvents. The physicochemical properties (matrix surface pattern, apparent matrix density, melting temperatures and swelling behavior) of the collagen materials and matrices were investigated. Furthermore, the diffusion coefficients of water inside the collagen devices (5.76*E-02cm²/h) and of various FITC dextrans in solution (e.g. FITC dextran 70: 2.4*E-03cm²/h) were determined by PFG-NMR and FCS, respectively. The developed collagen devices were used to investigate the enzymatic collagen matrix degradation and the release of higher molecular weight drugs, e.g. proteins. Several processes, i.e. diffusion, swelling and erosion, contribute to the overall release profile from collagen devices. Since it was desired to obtain a delivery system which controls release mainly by erosion, insoluble collagen type I materials were used to enhance the resistance against enzymatic attack. Besides this, collagen was physically or chemically cross-linked in some experiments to further restrict collagen digestion and drug delivery. It was shown that model compounds like BSA or FITC dextran 20, 70 and 150, respectively, could be incorporated and that their delivery could be controlled by the used collagen matrix material, e.g. animal source or cross-linking degree, the matrix dimensions (length or diameter of the extrudates), the molecular weight of the incorporated model compound and the drug load. The in vitro release of FITC dextrans and BSA was investigated and delivery of 80% model drug was in the range between 7h and 5d. Comparsion of the in vitro and the in vivo release (monitored in adult domestic pigs) of BSA was made by ESR. Similar results were obtained and it was shown that the mechanism of release changed from mainly diffusion towards erosion control by increasing the degree of matrix cross-linking. The degradation of insoluble collagen type I by bacterial collagenase was studied in detail to gain further insights into the enzymatic hydrolysis of collagen. In contrast to a simple Michaelis-Menten kinetic, adsorption of collagenase onto the substrate surface plays an important role. Based on the obtained in vitro results a mathematical model was developed to describe drug release from collagen matrices undergoing enzymatic degradation. Equations for the collagen degradation and the drug release were implemented, adsorption and diffusion phenomena were incorporated and a mixture of experimentally determined and fitted parameters was used to feed the model. Good correlation between experimental and simulated data was found. Histological evaluations demonstrated that the developed minirods showed good biocompatibility, with only minor inflammation reactions and normal tissue remodeling. This emphasized the assumption that collagen extrudates could be used in vivo without surgical removal after drug depletion.
