Abstract Participation of Neospora caninum in abortions in cattle populations in Northern Bavaria Stated aim of this study was to investigate the incidence and the potential importance of Neospora caninum as a cause of abortion in cattle populations in Northern Bavaria. For this purpose the available diagnostic methods of the LGL in Erlangen were compared with each other relating to their efficiency. Aim was to find out which method or which combination of methods is most useful in the routine diagnosis. To serve this purpose, all abortion cases which were sent in for analysis to the LGL Erlangen (232 cases) within a year (2005-2006) were tested with different direct and indirect methods. Direct diagnosis of the infection was determined by histopathology, immunohistochemistry and PCR (polymerase chain reaction). ELISA (Enzyme Linked ImmunoSorbent Assay) and Western Blot were used for indirect analysis. 23 of the 232 cases were considered to be infected by at least one of the diagnostic techniques used. That implies that Neospora caninum is involved in 10% of abortions in Northern Bavaria. It’s impossible to prove beyond doubt if Neospora caninum is the main cause of abortions or an innovator for further infections. Suspicious cases could be identified by routine histopathology because of characteristic lesions – particularly in brain and heart. Typical histological lesions consisted overbalancing in multifocal nonsuppurative necrotizing encephalitis and non-purulent myocarditis. Most of these cases were confirmed by immunohistochemical staining with using a polyclonal antibody. Frequently low numbers of disease agents necessitate more (up to 4) immunohistochemical runs in some cases to verify the agent. Especially in cases of bold autolysis or mummified fetuses histology was applicable only with restrictons, Immunohistochemistry not at all. PCR is able to detect DNA of the parasit in these cases. Altogether the PCR got the most positive results. In cause of this three different techniques (conventionell PCR, Real Time SYBR Green, Real Time TaqMan) could adduce approximately same results. The Real Time TaqMan Technique gained slightly advantage over the two other methods. High costs and difficulties to take positive results are detriments of the PCR-method. There is no qualified statement about the importance of Neospora caninum as a cause of abortion, using the PCR method only. Therefore the PCR method should be combined with one additionally morphological method. Serological tests of Neospora caninum intrinsic antibodies in fetal fluids proved not to be very useful. Fetuses in early pregnancy develop partly no specific antibodies against Neospora caninum so that these cases were seronegative in spite of showing typical morphological lesions. Compared with this, 4 seropositive cases did not show any histopathological alterations. A final interpretation of such cases is impossible. Summarising this study confirms the importance of Neospora caninum as a cause of abortion in Northern Bavaria and underlines the need to use different diagnostic techniques to increase the chance to detect the infection in aborted fetuses. The best combination is histopathology with PCR. After a screening with histopathology, suspicious fetal tissues should be verified by PCR. Furthermore the PCR has got the highest sensitivity of all used methods. It is able to detect also cases without lesions although inspite of presenting the parasit. PCR was confirmed to be the best tool for the diagnosis of Neospora caninum in aborted fetuses.

Influence of repetitive duodenal application of activated charcoal on the elimination of the elimination of intravenously applied acetaminophen (N-Acetyl-para-aminophenol) The influence of the repeated administration of activated charcoal on elimination of intravenously injected acetaminophen was investigated in an in-vivo model of anaesthetized rats. 40 rats were randomized and divided into four groups of 10 animals, respectively. Concentration of 14C-marked acetaminophen and its metabolites was measured in plasma, urine and small intestine irrigation samples of all animals. To measure the influence of activated charcoal on elimination of acetaminophen, the small intestine of one half of the animals (n=20) was perfused with activated charcoal dissolved in polyethylene glycol (PEG) (= gastrointestinal dialysis), the small intestine of the other half (n=20) was perfused just with PEG. In order to answer the question whether activated charcoal interrupts enterohepatic circulation, half of the rats treated with activated charcoal and half of the PEG-treated animals (n=10, respectively) were subject to bile duct cannulation; the externalized bile was then quantified for acetaminophen. During a testing period of 3.5 hours, in the ileal effluent of animals with physiological bile flow, we detected ca. 20% of the dose administered originally; in the animals subject to cannulation, we found about 7%. 13% of acetaminophen and metabolites were found in the externalized bile. Activated charcoal did not influence the exsorption of acetaminophen into the small intestine. Terminal half-life in blood ranged from 35-51 minutes, there was no statistically significant difference between the groups (P=0.152). Neither did the Area under curve (AUC) – ranging from 2.6 to 3.3 g/min./l. – show significant variation between groups (P=0.392). Concentration of acetaminophen in liver and kidney samples, which were removed post mortem, was very low, ranging from 0.02 to 0.6% of the dose originally administered. Excretion of acetaminophen into urine varied widely (31-56%), correlating with diuresis. The absence of an effect of activated charcoal on elimination of acetaminophen and metabolites may have been caused by exsorption of insufficient amounts into the intestinal lumen.

The prognosis for cats with fibrosarcoma is still poor as the treatment options existing to date do not lead to satisfying results. After sole surgical removal of the tumor, up to 70 % of the cats develop local recurrences. Even with adjuvant radiation and/or chemotherapy, the recurrence rate can just be reduced to at most 40 %. In the present work an alternative treatment method in terms of neoadjuvant immunostimulatory gene therapy should be established. Via plasmids, three feline cytokine genes were transferred into the tumor. The plasmids coded for feIL-2, feIFN-γ and feGM-CSF. Using magnetofection, gene transfer should be optimized. The aim of this clinical dose escalation study was to define a well tolerated dose, which can be tested for its efficacy in a subsequent phase-II trial. Therefore the cats were examined at defined time points for treatment-related toxicity up to 180 days after surgery. Two more check-ups on day 270 and 360 after surgery were performed to elongate the observation period for local recurrences. Prerequisite for a cat’s admission to the study was the localization of the fibrosarcoma (primary tumor or recurrence) on the trunk. The tumor had to be excised in one setting without limb amputation. Affected cats were neither allowed to be pretreated with chemo-, radiation- or gene therapy nor with corticosteroids within the past six weeks. Further exclusion criteria were hints for metastases or other severe illnesses which reduce life expectancy to less than one year. Due to the potential teratogenic effect of the expressed cytokines, pregnancy had to be ruled out. Only cats with histopathologically confirmed fibrosarcoma continued the study after surgery. As this was a scientific study with a drug not registered yet, written informed consent from the owners was a prerequisite for the participation of each cat. Four treatment groups with defined dose escalation were prospectively fixed. The dose of the feline cytokines was 15, 50, 150 and 450 µg per plasmid in group I, II, III and IV. The initial dose (3 x 15 µg) was oriented to the total dose for small oral melanomas in dogs (400 µg) established by DOW et al. and is 1/10 of it. Plasmids as non-viral vectors were chosen for several reasons. Their handling is not liable to such strict regulations as the potentially more dangerous viral vectors. Their production is simple and affordable. They induce fewer side effects than viral vectors. Therefore plasmids are more suitable for application in veterinary clinical practice. Equal amounts of the plasmids were brought into 0.9 % saline. The positively charged magnetic nanoparticles were brought into solution with aqua for injections and were mixed with the plasmid formulation in a 1:1 ratio. This formulation had a total volume of 500 µl and was injected twice intratumorally in weekly intervals. Transfection was enhanced and targeted to the tumor area by the application of a neodymium-iron-boron magnet for the duration of 60 minutes. One week after the second application, wide en-bloc resection of the tumor was performed. Four cats were assigned to each treatment group. As questionable toxicity occured in group IV, four more cats were added to this group. A control group also consisting of four cats received surgery without neoadjuvant therapy. For ethical reasons, the application of empty plasmids was avoided so that in this group surgery could be performed without delay. Medical care of all the cats was carried out by the same team of internists, anesthetists and surgeons. Clinical and laboratory parameters were evaluated according to the VCOG-CTCAE system. All adverse events were registered, classified with severity grades and correlated to treatment. Plasma samples of all cats up to 14 days after surgery were examined for the existence of feGM-CSF and feIFN-γ with commercially available ELISA kits. Statistical analyses were performed comparing the treatment groups themselves as well as treatment groups and the control group regarding body weight, white blood cells and differential blood counts. Only one cat out of group IV showed adverse events during the neoadjuvant treatment period, which were classified as grade 3 and which were probably correlated to treatment (correlation grade 4). For this reason four more cats were treated with the highest dose. None of these cats showed side effects that could be correlate to treatment. The occurrence of early recurrences in four cats of group IV was outstanding, but of course, the expressiveness of this statement is low regarding the small group sizes. However it is known that biological drugs, especially IL-2, often do not have a linear dose-response profile. Therefore the optimal effective dose lies within a strictly defined area and is probably lower than the maximal tolerated dose. The highest applied dose which is 450 µg per plasmid was defined as a well tolerated dose. It can be safely tested for its efficacy in a subsequent phase-II trial. Because of the reflections mentioned above it would undoubtedly be convenient to test the third dose (150 µg per plasmid) in parallel for its effectiveness.

A field study on lighting in the production of meat-type-chickens was conducted between April 2006 and February 2007. Part 1 comprised a survey on lighting practices in a substantial number of broiler farms in southern Germany (n = 187 broiler stables covering a total area of 276.337 m²). It was carried out on the basis of a questionnaire adapted to broiler management. The focus of data collection was put on animal welfare issues of lighting. It was found that in conventional broiler farming closed buildings, with or without windows, are the most common kind of facility used. This applies to the number of farms as well as to the total area. A comparatively high proportion of stables from the early days of poultry farming in Germany shows that these agricultural buildings often remain in use unchanged for decades. Thus, once taken, decisions on lighting have practical consequences over the whole life-cycle of an agricultural building. The surroundings in closed stables are primarily conceived to accommodate the need of farm staff for work-efficiency and workplace-safety. Scientific findings on visual perception of poultry have not been implemented systematically in poultry farming, yet even though lighting constitutes a central management factor which can influence behaviour and health of the animals to a substantial degree. An exception is the fact that conventional fluorescent lights with a flicker frequency of 100 Hz, which poultry may perceive as discontinuous, have largely been eliminated from the farms we examined. Subsequently the lighting environments of characteristic types of broiler sheds were examined by means of spectrophotometry. Data were collected on illuminance, irradiance, UVA-irradiance and colour temperature in summer as well as in winter. Substantial differences were found between the two sampling dates, particularly where ultraviolet radiation was concerned. Additionally considerable differences between the type of building (with or without daylight) could be described. Taking into consideration the enlarged sensory facilities of birds (perception of ultraviolet radiation, flicker fusion frequency higher than 100 Hz), it can be concluded that the currently used methods for sampling light intensity, particularly the lux-unit, are suitable for poultry to a limited extent only. Part 2 was a study on the effects of two lighting programmes which differed exclusively in intensity (5 lx and 20 lx) from the 13th day of the fattening period. The following parameters were collected and evaluated under field conditions: Development of intraocular pressure Development of eye measures Behaviour and time-budget Excretion of glucocorticoids (corticosterone) Performance parameters Under 5 lx a modulation of intraocular pressure was observed. Moreover all eye measures were larger under 5 lx than under 20 lx. A biological relevance of these differences, which were very slight, can be denied with a high certainty. The animals kept under 20 lx showed phases of higher activity and more explorative and comfort behaviour than the group raised under 5 lx. These results could be interpreted as a decrease in global activity due to the low light intensity. The lighting programme with an intensity of 5 lx resulted in a tendency for a higher excretion of fecal corticosterone, which was significant on day 35 only. This could be interpreted as a direct influence of dark surroundings on glucocorticoid metabolism. Bright lighting (20 lx) increased the risk for necrotic dermatitis (inflammatory process) by a factor of more than three. A prolonged fattening time of 37-39 days predisposed the animals to the development of necrotic dermatitis as well. Pododermatitis score was better under dim lighting. Further effects of lighting on production parameters could not be found. Taken together, these results give strong hints that a decrease of light intensity to approximately 5 lx at the beginning of the third week of a production cycle may constitute a tool to improve performance and prevent skin damage. Nonetheless these low light intensities could be stressful for the animals. These results were collected under field conditions and under certain restrictions. More research under controlled conditions remains therefore to be done.
