Molecular recognition, programmability, self-assembling capabilites and biocompatibility are unique features of DNA. The basic approach of DNA nanotechnology is to exploit these properties in order to fabricate novel materials and structures on the nanometer scale. This cumulative dissertation deals with three aspects of this young research area: fast analysis, autonomous control of functional structures, and biocompatible autonomous delivery systems for nanoscale objects. 1. At low temperatures and under favorable buffer conditions, two complementary DNA strands will form a double-helical structure in which the bases of the two strands are paired according to the Watson-Crick rules: adenine bases bind with thymine bases, guanine bases with cytosine bases. The melting temperature TM of a DNA duplex is defined as the temperature at which half of the double strands are separated into single strands. The melting temperature can be calculated for DNA strands of known sequences under standard conditions. However, it has to be determined experimentally for strands of unknown sequences and for applications under extreme buffer conditions. A method for fast and reliable determination of DNA melting temperatures has been developed. Stable gradients of the denaturing agent formamide were generated by means of diffusion in a microfluidic setup. Formamide lowers the melting temperature of DNA and a given formamide concentration can be mapped to a corresponding virtual temperature along the formamide gradient. Differences in the length of complementary sequences of only one nucleotide as well as a single nucleotide mismatch can be detected with this method, which is of great interest for the detection of sequence mutations or variations such as single nucleotide polymorphisms (SNPs). 2. Knowledge of the stability of DNA duplexes is also of great importance for the construction of DNA-based nanostructures and devices. Conformational changes occuring in artificially generated DNA structures can be used to produce motion on the nanometer scale. Usually, DNA devices are driven by the manual addition of fuel molecules or by the periodic variation of buffer conditions. One prominent example of such a conformational change is the formation of the so-called i-motif, which is a folded four-stranded DNA structure characterized by noncanonical hemiprotonated cytosine-cytosine base-pairs. In order to achieve controlled autonomous motion, the oscillating pH-value of a chemical oscillator has been employed to drive the i-motif periodically through its conformational states. The experiments were conducted with the DNA switch in solution and attached to a solid substrate and constitute the first example of DNA-based devices driven autonomously by a chemical non-equilibrium reaction. 3. Finally, a DNA-crosslinked and switchable polyacrylamide hydrogel is introduced, which is used to trap and release fluorescent colloidal quantum dots in response to externally applied programmable DNA signal strands. Trapping and release of the nanoparticles is demonstrated by studying their diffusion properties using single molecule fluorescence microscopy, single particle tracking and fluorescence correlation spectroscopy. Due to the biocompatibility of the polymerized acrylamide and the crosslinking DNA strands, such gels could find application in the context of controlled drug delivery, where the autonomous release of a drug-carrying nanoparticle could be triggered by naturally occurring, potentially disease-related DNA or RNA strands.

Over the last years, weather services have developed a new generation of high resolution mesoscale numerical weather prediction (NWP) models with the aim to explicitly predict convection. New methods are required to validate the representation of precipitation processes in these NWP models against observations. Polarimetric radar systems are especially suited for model validation as they provide information on the intensity and the microphysical characteristics of a precipitation event at a high temporal and spatial resolution. However, the observations can not be directly employed for model evaluation as polarimetric radar systems do not explicitly measure the parameters represented in microphysical parameterization schemes. In order to establish a relationship and allow for a direct comparison between the model parameters and the observations, the polarimetric radar forward operator SynPolRad (Synthetic Polarimetric Radar) has been developed. SynPolRad simulates synthetic polarimetric radar quantities out of model forecasts which permits an evaluation in terms of observed quantities. In a first step, the synthetic reflectivity, LDR, and ZDR are computed from predicted bulk water quantities and in a second step, the beam propagation in the model domain is simulated under consideration of refractivity and attenuation effects. In order to successfully employ SynPolRad for model evaluation purposes, the link between the forward operator and the mesoscale model has to conform as closely as possible to the model assumptions. However, in the case of a polarimetric radar forward operator not all the input parameters are defined by the model. Within this work, these free parameters are derived on theoretical terms accordingly to the model assumptions such that the polarimetric quantities match the thresholds of a hydrometeor classification scheme. Furthermore, special care is given to the representation of brightband signatures. The application of SynPolRad on two case studies proves the potential of the new method. A stratiform and a convective case study are chosen to assess the ability of mesoscale models to represent precipitation in different dynamical regimes. LMK (Lokal-Modell-Kürzestfrist) and MesoNH (Mesoscale Non-Hydrostatic Model) simulations considering different microphysical parameterization schemes are evaluated. The evaluation concentrates on the representation of life cycle, intensity, and the spatial distribution of synthetic reflectivity, LDR, and ZDR. Furthermore, hydrometeor types derived from the observed and synthetic polarimetric quantities employing a classification scheme are compared. Large discrepancies are found between the model simulations and the observations. However, the consideration of an additional ice hydrometeor category in the 3 component scheme significantly improves the performance of the LMK.

In the semi-analytical work presented here the feedback from supermassive black holes on galaxy clusters is investigated. In particular we aim at providing simple diagnostics tools to constrain the characteristic velocities and spatial scales of the hot Intra Cluster Medium (ICM) motions. In the so-called "cold core'' clusters these motions are believed to be driven by the activity of a central black hole. The methods developed here, together with present-day and future observations, are designed to help to solve the puzzle of cooling flow clusters (see section $1.3$) and understand better the AGN/gas interaction in smaller systems (down to individual galaxies).\\ Clusters of galaxies are the largest gravitationally bound systems in the Universe: they are composed of hundreds to thousands of galaxies, moving in a deep potential well set by the dominating dark matter. The whole volume of clusters is filled with hot (temperature $\sim 10^7-10^8$~K) and rarefied (electron density $10^{-4}-10^{-1} {\rm cm^{-3}}$) gas. In such a high-temperature regime even heavy elements (e.g. silicon, sulfur, iron etc.) are highly ionized up to [H]- or [He]-like ions and they emit in bright lines with energies from $\sim 0.7$ to $\sim 8$ keV. Using X-ray observations one can reliable measure all the major gas properties: the temperature, density and abundance of heavy elements.\\ A significant fraction of clusters (called "cool core'' clusters) show distinct signatures in the central region: the gas temperature drops inward, while the gas density increases. The central gas radiative cooling time in such clusters is much shorter than the age of the cluster and without any external source of energy the gas would cool well below X-ray temperatures. However observations suggest that the gas temperature drops only to 1-2 keV. One plausible explanation of this problem is that the activity of a central supermassive black hole deposits large amounts of mechanical energy into the cluster gas and that this balances the gas radiative losses. A direct implication of this hypothesis is that the hot gas is not at rest, but it is continuously stirred by the AGN activity.\\ The same class of cool core clusters is characterized by a centrally peaked distribution of the heavy elements abundance (usually measured using the He-like iron 6.7 keV line). The peaked abundance profiles are likely associated with the metals ejection by the stars of very massive elliptical galaxies, that are always present at the centers of these clusters. However the observed abundance distributions are significantly broader than the central galaxy light profiles, suggesting that some gas motions are spreading the metals ejected from the galaxy. We treat this process in a diffusion approximation to derive, from the X-ray observations, constraints on the characteristic velocities and spatial scales of the gas motions for a sample of cool core clusters and groups (Chapters $2$ and $3$). The parameters derived from a simple semi-analytic model are then compared with the results of numerical simulations of the AGN/gas interaction in the cluster core (Chapter $4$).\\ In Chapter $5$ we discuss the impact of the gas motions on the width of the strongest X-ray emission lines. Since the characteristic thermal velocities of heavy ions (e.g. iron) are much smaller than the sound speed of the gas, the width of the lines sensitively depends on the presence of gas motions. We show that both the absolute value of the linewidth and its dependence on the projected distance from the cluster center provide valuable diagnostics of the gas motions. Such measurements will soon become possible with the launch of X-ray micro-calorimeters in space.\\ This work has been done in collaboration with E.Churazov, R.Sunyaev, H.B\"ohringer, M.Br\"uggen, W.Forman and E.Roediger.

Abell 1689 is a massive galaxy cluster that has an impressive number of gravitationally lensed background galaxies. In this thesis more than 30 multiple image systems identified in A1689 are used to derive an accurate mass profile for the cluster. It is also shown how strong lensing in cluster can be used to constrain the sizes of the dark matter haloes of cluster galaxies and to place upper limits on the amount of dark substructure in clusters.

Absolute phase control of intense few-cycle pulses and steering the atomic-scale motion of electrons
1 Minute
In the past few years, ultrafast laser technology has developed to such a degree that the phase of a pulse under its envelope is now a meaningful measurable quantity. Many experiments now require the use of pulses with a fixed phase. The reliable production of such pulses, over extended periods of time, is of key importance to many areas of science. The central theme of this thesis is the generation of intense phase-controlled few-cycle optical pulses and their applications. This thesis reports on substantial improvements made in the generation of carrier-envelope phase-controlled pulses. Measurements performed show the accuracy to which absolute phase can be controlled has been improved to a unpreceded level. Also, the period of time over which such high accuracy measurements could be performed was extended by more than a factor of five, such that carrier-envelope phase-sensitive measurements that take longer than 24 hours without any breaks are now possible. With these tools at hand, physical processes that take place on sub-femtosecond time scales can be precisely measured, and control over the motion of bound electrons is possible. In this thesis, a report on the first demonstration of the latter is presented: In the dissociation of the D_2^+ -ion, control over the localisation of the remaining electron is demonstrated.

The analysis presented in this thesis is based on data from electron-proton collisions with longitudinally polarised electron beams at a centre-of-mass energy of ${\sqrt s}=319~$GeV. The data were taken with the H1 detector at the HERA collider in the year 2005 corresponding to two polarisation states: a left-handed electron polarisation of $-27\%$ and a right-handed electron polarisation of $+37\%$, corresponding to integrated luminosities of $68.6~\rm pb^{-1}$ and $29.6~\rm pb^{-1}$, respectively. This thesis presents inclusive $e^\pm p$ single and double differential cross sections for neutral current deep inelastic scattering measured as functions of the four-momentum transfer squared $Q^2$ and the Bjorken variable $x$ in interactions of longitudinally polarised leptons with unpolarised protons using the H1 detector at HERA~II. An overview of the phenomenology of deep inelastic scattering is given and the experimental apparatus as well as the measurement and analysis procedures are described. The cross sections are measured in the range of $200 < Q^2 < 20\,000$~GeV$^2$ and $0.0032
